fa شناسایی ژن‌های هاب کلیدی مرتبط با سلول‌های بنیادی سرطان سینه به ‌منظور غلبه بر مقاومت به درمان Identification of Key Hub Genes Associated with Breast Cancer Stem Cells to Overcome Therapy Resistance ژنتیک پزشکی medical Genetics پژوهشي Original <div style="text-align: justify;">زمینه: سرطان سینه یکی از شایع‌ترین و کشنده‌ترین بدخیمی‌ها در زنان است. حضور سلول‌های بنیادی سرطانی(CSCs) ، که در متاستاز، مقاومت درمانی و عود بیماری نقش کلیدی دارند، چالشی اساسی در درمان محسوب می‌شود. شناسایی اهداف درمانی اختصاصی مرتبط با CSCs از طریق رویکردهای زیست‌شناسی سیستمی می‌تواند به استراتژی‌های درمانی مؤثرتر منجر شود. مواد و روش‌ها: سه مجموعه ‌داده بیانی مربوط به سرطان سینه ) GSE7513سلول‌های بنیادی/ غیربنیادی سرطان(،GSE15852  (بافت‌های توموری/ نرمال)، و GSE76540 (سلول‌های مقاوم/ حساس به دوکسوروبیسین) از پایگاه GEO دریافت شد. تحلیل داده‌ها با ابزارهایGEO2R ، DAVID،STRING  و Cytoscape انجام گرفت. ژن‌های کلیدی بر اساس معیارهای شبکه‌ انتخاب (دیتاست‌های GSE7513 و GSE76540) و با ژن‌های متفاوت بیان‌ شده در GSE76540 مقایسه شدند. همچنین، بیان ژن‌ها، پیش‌بینی پاسخ به درمان و تعاملات دارو- ژن با ابزارهای TNMplot، ROC Plotter و DGIdb بررسی شد. یافته‌ها: داده‌های بیانی استخراج‌شده از مجموعه ‌داده‌ها به‌ترتیب 1446، 344 و 1826 ژن را شامل شدند. تحلیل اشتراک ژن‌ها بین GSE7513 و GSE15852، 75 ژن مشترک را شناسایی کرد. خروجی تحلیل شبکه برهمکنش پروتئین- پروتئین، 3 کلاستر و7 هاب ژن بود که 4 مورد آن با ژن‌های دارای بیان متفاوت در GSE76540 مشترک بود. ژن‌هایAGR2 ، GATA3 و KRT19 بر اساس TNM انتخاب شدند.KRT19  و GATA3با پاسخ بیماران به درمان ارتباط داشتند و بر اساس DGIdb، GATA3 به صورت بالقوه می‌تواند هدف داروهای موجود باشد. نتیجه‌گیری: این مطالعه نشان می‌دهد که GATA3 می‌تواند به‌‌عنوان یک نشانگر زیستی برای پیش‌بینی پاسخ به درمان و هدف بالقوه درمانی سرطان سینه مطرح باشد. </div> <div style="text-align: justify;">Background: Breast cancer is one of the most common and fatal malignancies in women. The presence of cancer stem cells (CSCs), which play a crucial role in metastasis, therapeutic resistance, and disease recurrence, presents a major treatment challenge. Identifying CSCs-specific therapeutic targets through systems biology approaches can lead to more effective treatment strategies Materials and Methods: Three gene expression datasets related to breast cancer— GSE7513 (Cancer stem/non-stem cells), GSE15852 (tumor/normal tissues), and GSE76540 (doxorubicin resistant/sensitive cells)— were obtained from the GEO database. Data were analyzed using GEO2R, DAVID, STRING, and Cytoscape. Hub genes were identified based on network parameters (GSE7513 and GSE15852 datasets) and compared with differentially expressed genes in GSE76540. Additionally, gene expression levels, treatment response prediction, and drug–gene interactions were evaluated using TNMplot, ROC Plotter, and DGIdb. Results: Gene expression data from the datasets included 1446, 344, and 1826 DEGs in GSE7513, GSE15852, and GSE76540, respectively. Comparative analysis between GSE7513 and GSE15852 identified 75 common DEGs. Protein– protein interaction network analysis output was three clusters and seven hub genes, four of them overlapped with DEGs in GSE76540. AGR2, GATA3, and KRT19 genes were selected based on TNM. KRT19 and GATA3 were associated with patients' response to treatment. According to the DGIdb, GATA3 is a potentially druggable target with existing therapeutic compounds.
