زمینه: هلیکوباکتر پیلوری شایع‌ترین باکتری است که جوامع انسانی را در ابعاد جهانی مبتلا به عفونت ساخته است. بیان فلاژل و تحرک باکتری در کلونیزاسیون و ویرولانس آن عامل بسیار مهمی است. ژن flaA یکی از ژن‌های کد کننده فلاژلین می‌باشد که نقش کلیدی در حرکت باکتری و کلونیزاسیون آن دارد و از نظر ایمنی بخشی نیز مورد توجه می‌باشد. هدف از این مطالعه طراحی، ساخت سازواره و بررسی بیان ژن flaA هلیکوباکتر پیلوری در سلول‌های یوکاریوتی است.

موادوروش‌ها: در این مطالعه تجربی، ابتدا از هلیکوباکتر پیلوری سویه استاندارد، DNA ژنومی تخلیص و ژن flaA با پرایمرهای اختصاصی به روش PCR تکثیر و جداسازی شد. سپس با بهره‌گیری از تکنیک همسانه سازی T/A در پلاسمید pTZ کلون گردید. به منظور بیان ژن flaA و ایجاد سازواره نهایی، ژن flaA از پلاسمید pTZ خارج شد و در وکتور بیانی (-)pcDNA3.1 ساب کلون گردید. سازواره -flaA(-)pcDNA3.1 به روش الکتروپوریشن به سلول جانوری CHO منتقل و بیان یوکاریوتی ژن flaA با تکنیک
SDS-PAGE بررسی شد.

یافته‌ها: نتایج نشان می‌دهد محصول PCR ژن flaA در وکتور pTZ کلون و در باکتری اشریشیا کلای سویه TOP10F تکثیر یافته است. همچنین هضم آنزیمی و تعیین توالی حاکی از تشکیل سازواره نهایی -flaA(-)pcDNA3.1 است. در نهایت بررسی بیان ژن flaA هلیکوباکتر پیلوری در سلول جانوری CHO، نشان داد سازواره ژنی حاصل قادر به بیان موفق محصول ژن flaA در سیستم یوکاریوتی می‌باشد.

نتیجه‌گیری: با توجه به اینکه سازواره نهایی -flaA(-)pcDNA3.1 قادر به بیان محصول پروتئینی ژن flaA هلیکوباکتر پیلوری در سلول‌های جانوری می‌باشد و پروتئین فلاژلین یکی از آنتی ژن‌های مهم این باکتری است. بنابراین به درستی می‌توان گفت که سازواره ژنی
-flaA(-)pcDNA3.1 کاندیدای مناسبی برای استفاده در زمینه واکسن‌های ژنی علیه هلیکوباکتر پیلوری می‌باشد و در تحقیقات آینده می‌توان از آن برای بررسی ایمنی زایی در حیوانات آزمایشگاهی بهره گرفت.
