زمینه: دو کفه‌ای‌های ماسل، پروتئین‎های چسبناکی تولید نموده که امکان اتصال موجود به سطوح در محیط‎های آبی را فراهم می‎کنند. خواص ویژه نظیر چسبندگی، مقاومت و سازگاری زیستی، کاربرد این پروتئین‎ها را در مهندسی پزشکی و دندان‌پزشکی و ساخت چسب‎های بیولوژیک مطرح نموده است.
مواد و روش‌ها: با توجه به شاخص‎های مورفولوژیکی و توالی ژن COI دوکفه‌ای ماسل (Modiolus sp. PG) از منشاء خلیج‌‌‌فارس نمونه‎گیری شد. به منظور تأئید حضور غده‎های پلی‎فنلی مولد مواد پروتئین‎ چسبنده، از بافت پای صدف مقاطع بافتی تهیه شد. سپس پروتئین fp-2 توسط روش‌های کروماتوگرافی تعویض یون با گرادیان نمک و pH و همچنین کروماتوگرافی ژل فیلتراسیون خالص‌سازی گردید. جهت اثبات وجود پروتئین‌ها در هر فراکشن از تست Arnow و رنگ‎آمیزی اختصاصی NBT استفاده گردید. جهت تشخیص خلوص پروتئین در هر مرحله از تخلیص از الکتروفورز SDS-PAGE استفاده شد. پیک‎های کروماتوگرام دارای پاسخ مثبت نسبت به NBT پس از تغلیظ و الکتروفورز (به روش اسید- اوره) آنگاه به کاغذ نیتروسلولز بلات و توسط رنگ‌آمیزی NBT تأیید گردید.
یافته‌ها: مشخصات گونه‎شناسی و ریخت‎شناسی صدف بررسی و با کلیدهای شناسایی FAO و کتاب نرم‎تنان سواحل خلیج‎فارس ایران تطبیق گردید. نمونه مورد مطالعه متعلق به خانواده Mytilidae و از جنس Modiolus بود. توالی نوکلئوتیدی و پروتئینی حاصل به ترتیب با شماره‎های AB972412.1 و BAP16437.1 در بانک اطلاعاتی NCBI ثبت گردید. همچنین وزن مولکولی پروتئین fp-2 تقریباً معادل 45 کیلو دالتون بود.
نتیجه‌گیری: این تحقیق منجر به شناسایی مورفولوژیکی و ژنتیکی صدف دوکفه‎ای خلیج ‎فارس (Modiolus sp. PG ) و تخلیص پروتئین چسبنده fp-2 گردید.
