زمینه: تب کیو (کوکسیلا بورنتی)، بیماری مشترک بین انسان و حیوان است و می‌تواند مشکلات جدی مانند نارسایی‌های کبدی، قلبی و عروقی، پنومونی و خستگی‌های مزمن طولانی را در میزبان ایجاد کند. با توجه به تأثیر مهم کوکسیلا بورنتی بر سلامت جامعه و محدودیت در استفاده از واکسن‌های طراحی شده قبلی، هدف از این مطالعه طراحی واکسن تب کیو بر اساس پروتئین‌های غشای خارجی کوکسیلا بورنتی با استفاده از روش‌های بیوانفورماتیک بوده است.
مواد و روش‌ها: اپی‌توپ‌های تحریک کننده لنفوسیت‌های B و T پروتئین‌های OMP1 و LolB با استفاده از سرورهای IEDB و ABCpred شناسایی شد. پس از ارزیابی ویژگی‌های مختلف مانند عدم سمیت (سرور ToxinPred)، عدم ایجاد آلرژی (سرور AllerTOP) و توانایی تحریک سیستم ایمنی (سرور VaxiJen)، اپی‌توپ‌های نهایی انتخاب و کاندید واکسن نوترکیب طراحی شد. ویژگیهای کاندید واکسن طراحی شده با استفاده از سرورهای مختلف (ProtParam، Solpro،PSIPRED ، Cluspro و C-ImmSim ) ارزیابی شد.
یافته‌ها: در نهایت، برای پروتئین‌ OMP1، 3 ‌اپی‌توپ تحریک کننده لنفوسیت B و 17 اپی‌توپ تحریک کننده لنفوسیت T شناسایی شد. همچنین 1 اپی‌توپ تحریک کننده لنفوسیت B و 9 اپی‌توپ تحریک کننده لنفوسیت T برای پروتئین‌ LolB شناسایی شد. ویژگی‌های مختلف کاندید واکسن طراحی شده مانند ویژگی‌های فیزیکوشیمیایی، ساختار ثانویه، عدم سمیت و توانایی تحریک سیستم ایمنی مثبت پیش بینی شد.
نتیجه‌گیری: بر اساس نتایج محاسباتی مطالعه حاضر، کاندید واکسن طراحی شده ممکن است به خوبی سیستم ایمنی را تحریک و موجب ایمنی در برابر بیماری تب کیو شود. جهت اطمینان از اثربخشی و ایمن بودن کاندید واکسن طراحی شده مطالعات تکمیلی در شرایط آزمایشگاهی و بدن موجود زنده ضروری است.
